Abstract: Arthritic degeneration of knee joint is one of the most common kinds of musculoskeletal disease in which cartilage degeneration takes place evidently. It confines the range of motion (RST OM) in affected joints, consequently decreasing the functional ability of the patient. Due to this reason arthritis is one of the leading causes of deaths due to inactivity. Keeping an eye on this fact that this study was planned to find out a new dimension regarding the management of this unkind ailment. The therapeutic associations between the exercises, ultrasound therapy and enzyme supplementation were investigated by a randomized control trial. It was conclude that all interventions but especially two of the included interventions (therapeutic ultrasound and enzymes supplementation), exhibit physiological and statistical considerable effects. The results were compared in two dimensions, one on the basis of physical parameters i.e. pain, range of motion, muscle strength and functional status and secondly on the basis of biochemical parameters i.e. ALP, SGPT, SGOT and Cholinesterase. The most significant component of this study is High Performance Liquid Chromatography analysis of human serum. HPLC chromatograms showed remarkable absorption of enzymes supplementation when sample were analysed pre-treatment and after treatment. After it was concluded that exercise can be suggested to the patients with mild to moderate arthritis and a combined therapy with enzyme supplementation can make, further improvement in daily living activities of patients with knee joint osteo arthritis.
